The p53 gene plays a key role in the prevention of cancer, by blocking cell growth and triggering programmed cell death or apoptosis. If, however, p53 has mutated and become defective, the cancer cells can acquire the ability to evade apoptosis and become more resistant to therapy. Researchers at Karolinska Institutet and Karolinska University Hospital in Sweden have now obtained results from the first tests using a new substance that can restore the function of defective p53 and activate apoptosis in cancer cells.

The substance is known as APR-246 and has now been tested on humans in a phase I/II study, which was conducted on 22 patients with advanced blood or prostate cancer. Some of the patients came from the Haematology Centre at the Karolinska University Hospital in, Stockholm, where the study's lead investigator, consultant Dr Sren Lehmann is based. The remainder of the patients were from other clinics in Gothenburg, Lund, Uppsala and rebro.

The patients received daily infusions of APR-246 for four days. When the researchers analyzed the cancer cells taken before and after treatment, they saw indications that the p53 gene had been activated to varying degrees, and that this had triggered the suicide program in the cancer cells. Ten patients could be evaluated as regards the development of their cancer, and in two of them there were signs of tumour regression.

However, the study was actually not designed to test the clinical effects but to ascertain how well the substance was tolerated by the body. With the main adverse reactions confined to temporary tiredness, nausea, headache and confusion, their results would suggest that the substance is well tolerated.

"The side-effects were totally different to those produced by conventional chemotherapy, which bodes well for designing combination therapies," says Dr Lehmann. "And it's in precisely this kind of combination that we think the substance has the greatest potential. In previous laboratory studies we've seen that APR-246 has generated synergy gains when used with chemotherapy due to the mutually enhancing effects of both substances."

Defective p53 is considered one of the most common factors behind the development of cancer. In some cancers, such as ovarian cancer, the vast majority of tumours have defective p53. In total, the p53 tumour suppressor gene is mutated in at least half of all tumours.

"In theory, a drug that restores p53 function should be effective against many different kinds of cancer, provided that the individual tumour contains defective p53," says study team member Professor Klas Wiman. "We should keep in mind, however, that tumours are very complex."

APR-246 was discovered by Klas Wiman and colleagues at Karolinska Institutet, and the present study was led from Karolinska University Hospital in association with Aprea AB. Aprea AB's principal shareholder is Karolinska Development, a company listed on the NASDAQ OMX Stockholm exchange. Professor Wiman is co-founder and shareholder of Aprea, and a member of its board.

99% Finding Nemo 3D

A genuinely funny and touching film that, in less than a decade, has established itself as a timeless classic.

It makes even more compelling what is still my all-time favorite Pixar film.

Think of this re-release as an encore, a handy touchstone for you and your kids. "Finding Nemo" was and remains the gold standard against which all other modern animated films are measured, a classic from the day it premiered.

In this seamless blending of technical brilliance and storytelling verve, the Pixar team has made something as marvelously soulful and innately, fluidly American as jazz.

Nemo, with its ravishing underwater fantasia, manages to trump the design glamour of earlier Pixar films.

A great blend of storytelling and technology.

It's downright curmudgeonly to remain focused on the negatives when the rest of the picture is saturated with invention and wit.

It's bright, it's beautiful and it hasn't aged a day. So why, then, do we need to see it in 3-D?

A rare example of a movie that not only survives its 3-D conversion but benefits from it

It was as funny as I remember and brilliant because it is subtle. It just knocks it out of the park. I love everything about this movie, one of my favorite Pixar movies of all time, 5/5 Schmoes. The new sound/3D adds to the greatness that is Nemo.

If you're fishing for fun in an ocean of movie mediocrity, grab the kids or the grandkids, fork out a few extra bucks and see this one in three-dimensions.

It's a great movie, so much fun, an acheivement that should be enjoyed by children of all ages and worth seeing it in 3D.

Sea-worthy w/enhanced visual lure and rich, humorous characters set in a double narrative; this 3D revisit allows us to fall overboard all over again.

If ever there was a film that wasn't broke, this is it. So naturally they converted it into 3D for a special re-release this weekend. (Stupid irony.)

I believe my sensitivity to certain creative choices of the picture has dulled some, while age has opened my eyes to its complex relationships and tireless sense of exploration.

When the blunt head of Bruce the shark slams into the screen like a hammer, everyone -- adults included -- gasps

At its best, the added dimension is merely harmless, making the movie blurry enough that you need special glasses to make it sharp again.

[VIDEO ESSAY] Disney/Pixar's 3D upgrade of its classic animated children's movie - using the expensive Xpand 3D format - has the duel effect of darkening the film's vibrant colors while adding considerable depth to the screen.

If the Pixar vault was on fire and they could only save one film, it'd be this eminently re-watchable modern fairy tale...Looks all the more stunning with this state-of-the-art 3D retro-fit.

I could write an entire dissertation on how much I love this movie.

Fading buzz from Fed may mean quiet market

NEW YORK -- Comparing the Federal Reserve to a rehab clinic offering addicted investors a synthetic high has been a favorite of Wall Street wags ever since the first round of Fed stimulus nearly four years ago. The punch line is that you always need more and more to get the same high and each bout of euphoria is followed by a crashing comedown.

After the frenetic reaction brought about by the announcement of the Fed's latest stimulus program -- $40 billion pumped into the U.S. economy each month -- the coming week is likely to bring a more sober period for markets as investors digest what it means in the longer run and turn their attention to the remainder of the year.

That will include rancorous U.S. elections in November, wrangling over taxes and spending cuts and a slowdown in corporate earnings.

"Right now we have this short-term euphoria. But then the question is where do we go from here," said Frank Fantozzi, chief executive of Planned Financial Services, an independent wealth manager in Cleveland. "I think after a week or so, if the underlying economic data doesn't change, you're going to see the market drop a bit and we'll continue to plod along until the election."

The effect on markets of the European Central Bank's plan to buy government bonds of struggling euro zone countries, then the Fed's opened-ended commitment to spur growth have been breathtaking. The Dow and the S&P 500 reached the highest closing level in nearly five years while the Nasdaq marched to new 12-year highs.

But in Friday's stock market action strong gains in the morning steadily eroded throughout the day, perhaps the first signs of fatigue creeping into the market.

"We are starting to get into that heady territory where you need to be on the defensive," said Richard Ross, global technical strategist at Aubach Grayson in New York. "Trying to squeak out the last 5 percent of a move when there is potentially a 15 to 20 percent downside in my opinion is pretty dangerous stuff."

Ross believes that equities, commodities and currencies are now approaching extreme levels of both price and momentum while geopolitical tensions in the Middle East are rising.

Even though all the major stock market rallies since the financial crisis have coincided with new central bank efforts to stimulate the economy, not everyone is buying it.

The latest data shows a moderate increase in short interest -- bets that stocks will fall -- across S&P 500 stocks during the last two weeks of August, a period when stocks were rallying on expectations of the Fed's announcement. Typically short interest inversely tracks the market. If investors were getting out of bets that stocks will fall, that would mean buying back those stocks and forcing the market higher.

Data provided by Schaeffer's Investment Research, a Cincinnati-based research firm, shows that bets against the biggest 500 U.S. companies edged back to about 7.3 billion shares after falling from about 7.6 billion to 7.2 billion from the start of July through the end of August -- a period when the market gained more than 3 percent.

That uptick in short interest could be significant. From the middle of September 2011 through the end of May this year, short interest on S&P 500 stocks fell like a stone to about 6 billion shares. During that period the S&P hit a four-year high, rising more than 20 percent from trough to peak.

One side effect of the Fed's bond-buying should be to reduce volatility in markets. That means the CBOE VIX volatility index?should remain close to the five-year lows it hit this summer. In August it fell as low as 13.30.

Yet activity in the options market shows some very bold bets that volatility could skyrocket in the months ahead. Call option buying on the VIX -- bets the index will rise -- is close to a record high at 5.182 million contracts, according to Schaeffer's data. The record is 5.249 million set in August.

The most actively traded VIX calls on the Chicago Board Options Exchange were October calls with a strike price of 60. Those also had the highest open interest. The VIX would need to rocket more than 300 percent by mid-October, hitting its highest level in about four years, for that trade to break even.

On the face of it a bet like that may seem little better than betting your 401(k) on a single number on the roulette table, but it does reveal more bearishness creeping into the market as stock indexes march to new highs.

Open interest, or outstanding contracts, on the October VIX calls with a strike price of 60 was 37,000 while traded volume was around 40,000. Many of those were bought in blocks of 2000 to 5000 for 5 cents each, suggesting a single buyer, according to Todd Salamone, vice president of research at Schaeffer's.

"Somebody's really playing a disaster by October," said Salamone. "If they're looking for something that big, that is not a portfolio hedge because that would be a lot of downside in the market before that hedge would actually kick in."

A lot of the market action over the coming months will depend on whether the Fed's stimulus program succeeds in boosting the economy.

Data on the housing market in the coming week is expected to show a continuing improvement. Economists in a Reuters poll expect the National Association of Home Builders Index to tick up in September when the data is released on Tuesday. On Wednesday both housing starts and existing-home sales are expected to increase.

Early manufacturing data for September, however, is not expected to be so robust. Both the Empire State index and the Philadelphia Fed index are tipped to show contraction. That would follow the sharpest drop in U.S. manufacturing in more than three years in August, which was also the third consecutive month of contraction.

"Let's face it -- we are in truly unchartered waters here," said Nicholas Colas, chief market strategist at the ConvergEx Group in New York.

Leinenkugel's new Lemon Berry Shandy was almost apple

The new Lemon Berry Shandy from Jacob Leinenkugel Brewing Co. was almost an apple shandy, Dick Leinenkugel recently told me.

Leinenkugel on Aug. 29 announced its second shandy offering, piggybacking off the success of the mega-popular Summer Shandy. Leinenkugel is a subsidiary of Milwaukee-based Tenth and Blake Beer Co., the craft and import division of Chicago-based MillerCoors LLC that has seen strong growth since its 2010 inception.

Several different flavors were tested before the company settled on lemon berry as a new, year-round shandy, said Dick Leinenkugel, business development manager for Tenth and Blake.

?I remember in March at the MillerCoors distributor conference in Las Vegas, we were sampling an apple shandy,? Leinenkugel told me. ?It was received interestingly, but I would say not enthusiastically. We made a very quick decision at that point to say, ?This is not optimal, we need to go back to the drawing board.??

The company went to work on what eventually become Lemon Berry Shandy, which Leinenkugel said scored equal to and in some cases higher in likability than Summer Shandy among test audiences.

?We knew then that we had a winner,? Leinenkugel said.

"Fly Me to The Moon" was written by Bart Howard in 1954. It was originally titled "In Other Words." Frank Sinatra's 1964 recording was played by the astronauts of Apollo 10 and it was played again on the moon by Buzz Aldrin during the Apollo 11 landing. And Diana Krall sang it in 2009 during the 40th anniversary of the landing, with all three of the Apollo 11 crew present at the Smithsonian.

Jennifer Lopez already has her own production company, Nuyorican Productions, and now the former "American idol" judge will also be part-owner of a cable TV network, the English-language, network for Latinos, nuvoTv.

With a growing Latino audience, there is more demand for television programming around that audience. Until now, there have only been a few networks focused on this demographic, and really none focused on providing Latino cultural content in English.

As Lopez told USA Today, when she was growing up there was very little programming that appealed to her heritage.

"Growing up in the Bronx as a little Puerto Rican girl watching TV and not having a lot to identify with. West Side Story was really my favorite thing because it had Puerto Ricans in it."

According to AdAge, Nuvo isn't the first television network to enlist the presence of celebrities to market its content and increase viewership. At least for Lopez, this was a natural step in the right direction.

"There is no English network that really focuses on our story," Lopez told AdAge. "I'm in this business and I do know this business on many different levels -- in music, in television, in film production, and as an actress, and in so many different ways, even in fashion. It's kind of right up my alley."

In August, nuvoTV announced it had raised $40 million to produce a new lineup of original English-language programming, illustrating "the gap in the market, along with the dire need to fill it with Latino TV shows for English-language speakers."

(AP) ? The number of Americans seeking unemployment benefits jumped to the highest level in two months, although the figures were skewed in part by Hurricane Isaac.

Applications increased by 15,000 to a seasonally adjusted 382,000, the Labor Department said Thursday. That's up from 367,000 the previous week. The four-week average, a less volatile measure, increased for the fourth straight week to 375,000.

Isaac made landfall as Category 1 hurricane on Aug. 28 in southeastern Louisiana and was later downgraded to a tropical storm. It disrupted work in nine states and boosted applications by roughly 9,000, Labor officials said.

Applications for unemployment benefits reflect the pace of layoffs.

The data on unemployment applications follow last week's disappointing August employment report. And it comes as the Federal Reserve concludes a two-day meeting. Many analysts expect the Fed will announce new steps to boost the economy when the meeting ends.

Jim O'Sullivan, chief U.S. economist at High Frequency Economics, said that after excluding the impact of the hurricane, applications are likely closer to 370,000. Still, the job market has shown little vigor this year.

"While today's reading was likely exaggerated, and we do not believe the labor market is weakening, the trend has been and still is weak enough to trigger more Fed easing today," O'Sullivan said.

Employers added only 96,000 jobs in August, below July's gain of 141,000 and far below the average 226,000 a month added in the January-March quarter.

The unemployment rate dropped to 8.1 percent from 8.3 percent, but only because the number of people working or looking for work fell.

The total number of people receiving unemployment aid fell to 5.4 million in the week ending August 25, the latest data available. That's about 80,000 fewer than the previous week. Some of the decline may result from more people getting jobs. But many recipients are using up all the benefits available to them.

Many economists have noted that while layoffs are falling, overall hiring isn't picking up at the same pace. A separate monthly report from the Labor Department this week showed that layoffs were at the lowest level in July in the 11 years the government has tracked the data.

The economy isn't growing fast enough to support much more hiring. It grew at a tepid 1.7 percent annual rate in the April-June quarter, down from 2 percent in the January-March quarter and 4.1 percent in the final three months of last year.

Growth isn't likely to get much better for the rest of this year. Economists expect it to grow at a roughly 2 percent pace. That's typically too weak to create enough jobs to lower the unemployment rate.

High unemployment and sluggish growth has prompted many economists to forecast that the Federal Reserve will announce new steps to boost the economy after its two-day meeting ends Thursday. Those steps could include a third round of Treasury bond purchases. The goal of the purchases would be to lower interest rates and spur more borrowing and spending.

Fed Chairman Ben Bernanke said in a recent speech that weak hiring is "a grave concern" that causes "enormous suffering."

First group of delayed young deportees approved

WASHINGTON (AP) ? Just three week after the Obama administration started accepting applications from young illegal immigrants seeking to avoid deportation and get a work permit, the government already has approved some of the roughly 72,000 applications the government has received.

The Homeland Security Department said Tuesday that a small group of applications has been approved and those immigrants are being notified this week about the decision. The department did not say how many applications had been approved.

The first wave of approvals comes months head of DHS' own internal estimates of how long the application process for the administration's Deferred Action for Childhood Arrivals program could take ? and less than 60 days before the Nov. 6 elections. According to an internal DHS document obtained by The Associated Press, the department's Citizenship and Immigration Services had estimated that each application could take several months to be completed.

"Following a thorough, individualized case review, USCIS has now begun notifying individuals of the determination on their deferral requests," DHS spokesman Peter Boogaard said in a statement. He said about 72,000 applications have been received since the program's Aug. 15 start.

President Barack Obama and Homeland Security Secretary Janet Napolitano announced on June 15 that young illegal immigrants who came to the U.S. before they turned 16, are 30 or younger, are high school graduates or are in college or have served in the military would be eligible to apply to avoid deportation for up to two years and get a work permit. The immigrants also could not have a serious criminal record.

The policy change came just months before what is shaping up to be a tight presidential election. Wooing Hispanic voters has been considered key to helping Obama win a second term.

The plan to halt deportations for as many as 1.7 million illegal immigrants closely mirrors the failed DREAM Act, a bill that would have provided a path to legalization for many of the same immigrants expected to benefit from the government' deferred action policy. The new policy does not provide legal status for the immigrants, but instead puts off possible deportation for up to two years.

Republican presidential nominee Mitt Romney has not said what he would do with Obama's policy if he is elected. He has previously pledged to veto the DREAM Act should it cross his desk. DREAM stands for Development, Relief and Education for Alien Minors.

Republicans have uniformly criticized Obama's policy, as well as previous DHS decisions to stop deporting many illegal immigrants who do not have criminal records or otherwise pose a threat to national security or public safety.

Rep. Lamar Smith, R-Texas, the chairman of the House Judiciary Committee, has derided the policy as "backdoor amnesty" and said he worried about fraud.

"President Obama and his administration once again have put partisan politics and illegal immigrants ahead of the rule of law and the American people," Smith has said previously.

Last month a group of Immigration and Customs Enforcement agents sued the department in federal court in Dallas and accused the administration violating federal law and forcing ICE employees to break the law by not arresting certain illegal immigrants. Napolitano and ICE Director John Morton are named as defendants.

An informal adviser to Romney on immigration, Kansas Secretary of State Kris Kobach, filed the suit on behalf of the agents.

"It places ICE agents in an untenable position where their political superiors are ordering them to violate federal law," Kobach said at the time. "If they follow federal law, they will be disciplined by their superiors."

On Tuesday, Sen. Jeff Session, R-Ala., sent a letter to Morton questioning the new policy and how it is being implemented. He cited reports of immigration officers being threatened with punishment for trying to enforce immigration laws, an allegation outlined in Kobach's suit.

"Your job is to carry out the mission of your agency and support the officers on the front lines," Sessions wrote. "Your apparent failure to support your officers in these incidents and your evident lack of concern for the administration's decision to nullify the very laws they were sworn to enforce raises serious questions about your ability to lead this agency."
